肝癌是中国高发恶性肿瘤。由于其广泛的肿瘤异质性，最新的靶向联合免疫治疗的方案仅对20~30%的患者有效，并且缺乏指导精准用药的药敏分子分型。因此亟需研究可以指导肝癌精准用药的分子分型和揭示新的耐药靶点。
Hippo信号通路在肿瘤发生发展中发挥重要作用，其效应蛋白YAP/TAZ转录辅助因子在多种肿瘤中显著激活[1, 2]。因此，靶向Hippo通路的肿瘤治疗策略具有很好的临床转化前景。然而，目前尚无相关药物真正进入临床用于肿瘤治疗[3]。近年来，基于抗体和细胞的靶向疗法方兴未艾，如果能鉴定出一种体现Hippo通路活性的细胞表面标志物，将为靶向Hippo通路的肿瘤治疗带来全新可能性。
Hippo信号通路参与肿瘤发生、发展、耐药等不同阶段，是一个理想的肿瘤治疗靶点。近年来，开发靶向Hippo通路的抗癌疗法已成为一个研究热点，但目前尚没有相关药物进入临床。明确Hippo通路调控机制是分子靶向治疗研发的重要基础。然而，Hippo领域近二十年的研究主要聚焦于通路组分的鉴定及功能解析，分子机制研究则相对滞后。
髓母细胞瘤是儿童最常见的颅脑恶性胚胎性肿瘤，世界范围的发病率为每年5.07-6.8/10万人，是引起儿童肿瘤相关死亡的首要原因。目前针对髓母细胞瘤的治疗是一种联合治疗的策略，手术为主，综合放疗以及化疗、靶向药物治疗等作为辅助治疗。髓母细胞瘤的发病机理极其复杂，涉及表观遗传学、胚胎发育学等多个方面，目前尚未完全明确，导致总体治疗效果并不是非常理想。
